1. Hanging drop method

Take a small amount of typical secretion, place it on a glass slide with warm saline or 10% potassium hydroxide solution, mix it well, and look for Trichomonas or vaginal Candida spores and pseudohyphae under a microscope.

2. Amine Odor Test

Take a small amount of vaginal secretion, put it on a glass slide, add 1-2 drops of 10% potassium hydroxide solution, and it is positive if a fishy smell like rotten fish is produced. This is due to the release of ammonia when amine meets alkali, which is a characteristic of bacterial vaginosis.

3. Clue Cells

Take a small amount of secretion and place it on a glass slide, add a drop of physiological saline and mix it. If clue cells are found under a high-power microscope, the diagnosis of bacterial vaginosis can be considered. Clue cells are surface cells that fall off the vagina. The granular substances attached to the edge of the cells are various anaerobic bacteria, especially Gardnerella, and the cell edges are unclear.

4. Cultivation Method

When the hanging drop method cannot confirm the diagnosis of a suspected patient, secretions can be taken for pathogen culture.
